大气PM_(2.5)对大鼠海马组织脑源性神经营养因子及其受体TrkB蛋白表达的影响Effect of atmospheric PM_(2.5) on the protein expression of brain-derived neurotrophic factor and its receptor TrkB in the hippocampus tissue of rats

摘要(Abstract):
目的 探讨大气PM_(2.5)染毒对大鼠海马组织中脑源性神经营养因子(BDNF)及其受体TrkB相关蛋白表达的影响。方法 采集大气PM_(2.5),制备混悬液。将48只SPF级雄性SD大鼠随机分为对照组和暴露组,每组8只,染毒组大鼠按照20 mg/kg气管滴注PM_(2.5)混悬液,对照组大鼠气管滴注等量生理盐水,每周1次,分别染毒1个月、3个月和6个月。采用免疫组化法测定大鼠海马组织中PKA蛋白表达,蛋白免疫印迹法测定大鼠海马组织中BDNF、TrkB、AKT和CREB蛋白表达水平。结果 与相应对照组比较,3个月、6个月染毒组大鼠海马组织中PKA阳性细胞减少,BDNF、TrkB、AKT和CREB蛋白表达水平均明显降低,差异均有统计学意义(P<0.05)。结论 大气PM_(2.5)染毒可降低大鼠海马组织中BDNF、TrkB、AKT、CREB和PKA蛋白表达水平。
